Product Specific InformationMA1-023-D488X has successfully been used in immunofluorescence assays with human samples.Target InformationTRA-1-60 is a cell surface antigen, expressed along with SSEA-3, SSEA-4 and TRA-1-81 in human embryonic stem cells, embryonal carcinoma cells and induced pluripotent stem cells (iPS). These surface markers are lost during the differentiation process. In contrast, SSEA-1 is absent in undifferentiated human stem cells but is present on the cell surface after retinoic acid mediated differentiation.For Research Use Only. TRA-1-60 Antibody (MA1-023-D488X) in IFImmunofluorescent analysis of TRA-1-60 (green) in induced pluripotent stem cell (iPS) clones from a regenerative medicine platform (anonymous). Cells were probed with a DyLight 488-conjugated TRA-1-60 monoclonal antibody (Product # MA1-023-D488X) at a dilution of 1:10. Nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken at 10X magnification. TRA-1-60 Antibody (MA1-023-D488X) in IFImunofluorescent analysis of TRA-1-60 (green) in human episomal iPSC cells. Cells were grown on vitronectin coated plates, fixed with 4% paraformaldehyde for 15 minutes, and blocked with 3% Blocker BSA (Product # 37525) for 30 minutes at room temperature. Cells were stained with DyLight 488-conjugated TRA-1-60 monoclonal antibody (left panels, Product # MA1-023-D488X) or DyLight 488-conjugated Mouse IgM isotype control antibody (top right panel, Product # MA1-194-D488), at a dilution of 1:250 and incubated for 1 hour at room temperature. Nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on an EVOS FL Imaging system at 20X magnification. TRA-1-60 Antibody (MA1-023-D488X) in FlowFlow cytometry analysis of TRA-1-81 in HEL 11.4 cells (blue histogram) compared to negative control unstained cells (red histogram). Cells were incubated with a DyLight 488-conjugated TRA-1-60 monoclonal antibody (Product # MA1-023-D488X) in 5% BSA-PBS at a dilution of 1:100 for 60 minutes for 1 hour on ice, washed and suspended into 500 µL of 5% BSA-PBS-buffer containing 10 µL 4% paraformaldehyde prior to FACS analysis.
