Product Specific InformationIntact IgG appears on a non-reducing gel as ~150 kDa band and upon reduction generating a ~25 kDa light chain band and a ~50 kDa heavy chain.Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems. The expression systems are developed by cloning in the specific antibody DNA sequences from immunoreactive rabbits. Then, individual clones are screened to select the best candidates for production. The advantages of using recombinant rabbit monoclonal antibodies include: better specificity and sensitivity, lot-to-lot consistency, animal origin-free formulations, and broader immunoreactivity to diverse targets due to larger rabbit immune repertoire.Target InformationThe c-Myc protein is a 62 kDa transcription factor that is encoded by the c-Myc gene on human chromosome 8q24. c-Myc is commonly activated in a variety of tumor cells and plays an important role in cellular proliferation, differentiation, apoptosis and cell cycle progression. The phosphorylation of c-Myc has been investigated and previous studies have suggested a functional association between phosphorylation at Thr58/Ser62 by glycogen synthase kinase 3, cyclin dependent kinase, ERK2 and C-Jun N terminal Kinase (JNK) in cell proliferation and cell cycle regulation. Studies have shown that c-Myc is essential for vasculogenesis and angiogenesis in tumor development, which distributes blood throughout the cells. The c-myc oncogene (p62 c-myc) is involved in the control of normal cellular proliferation and differentiation, and the deregulated expression of c-Myc induces apoptosis in different cell types. Antibodies against c-myc epitopes recognize overexpressed proteins containing Myc epitope tag fused to either amino- or carboxy-termini of targeted proteins. c-Myc-, N-Myc- and L-Myc-encoded proteins function in cell proliferation, differentiation and neoplastic disease. Amplification of the c-Myc gene has been found in several types of human tumors including lung, breast and colon carcinomas, while the N-Myc gene has been found amplified in neuroblastomas. Translocation of the c-myc locus on chromosome 8 to the immunoglobulin loci on chromosome 14 (heavy chain); 2 (delta light chain); or 22 (light chain) is described in Burkitts lymphoma and other B-cell lymphoproliferative conditions. An aberrant expression of the c-myc gene occurs in tumors of different origins such as colorectal, gastric, gallbladder, hepatic, mammary, ovarian, endometrial, head and neck, pulmonary, prostatic, thyroidal, oral, ocular, nasopharyngeal, endocrine, as well as hematopoietic neoplasms.For Research Use Only. c-Myc Antibody (700648) in IFImmunofluorescent analysis of c-Myc was performed on 70% confluent log phase U-2 OS cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0. 25% Triton™ X-100 for 10 minutes, and blocked with 5% BSA for 1 hour at room temperature. The cells were labeled with ABfinity™ c-Myc recombinant rabbit monoclonal antibody (Product # 700648) at a dilution of 1:1000 in 1% BSA and incubated for 3 hours at room temperature and then labeled with Alexa Fluor® 488 goat anti-rabbit IgG secondary antibody (Product # A-11008) at a dilution of 1:400 for 30 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938) and F-actin (Panel c: red) was stained with Alexa Fluor® 594 phalloidin (Product # A12381). Panel d is a merged image showing nuclear localization and panel e is a control without primary antibody. The images were captured using a Nikon microscope at 20X magnification.

c-Myc Antibody (700648) in WBWestern blot analysis of c-Myc was performed by loading 20 µg of Jurkat, A549, K562, U-87 MG, A431, HEK-293, and MDA-MB-231 cell lysates using Novex®NuPAGE®4-12% Bis-Tris gel (Product # NP0321BOX), XCell SureLock Electrophoresis System (Product # EI0002), Novex® Sharp Pre-Stained Protein Standard (Product # LC5800), and iBlot® Dry Blotting System (Product # IB21001). Proteins were transferred to a nitrocellulose membrane and blocked with 5% skim milk for 1 hour at room temperature. c-Myc was detected at ~57 kDa using ABfinity c-Myc recombinant rabbit monoclonal antibody (Product # 700648) at a 1:1000 dilution in 2.5% skim milk at 4°C overnight on a rocking platform. Detection was performed using an HRP-conjugated goat anti-rabbit secondary antibody (Product # G-21234) at a 1:5000 dilution and chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
