Classic vs fast Western protocol using SuperSignal West Pico Substrate293T cell lysate was diluted in reducing sample buffer and loaded to a gel. After electrophoresis, proteins were transferred to nitrocellulose and PVDF membranes using a semi-dry blotter for 10 minutes at 25V. Half of both membranes was developed using the classic Western protocol, and the other half was developed using the fast Western protocol. XIAP detection: The blots were probed with mouse anti-XIAP antibody (1µg/mL, BD Pharmingen), incubated in SuperSignal West Pico Substrate Working Solution for 5 minutes and exposed to film for 1 minute. TSC1 detection: The blots were probed with mouse anti-TSC1 antibody (1/1000 dilution, Cell Signaling Technology), incubated in SuperSignal West Pico Substrate Working Solution for 5 minutes and exposed to film for 5 minutes. |
