Serial dilutions of HeLa lysate (12.5μg and down) were separated by SDS-PAGE, transferred to nitrocellulose or PVDF membrane and then blocked for 5 minutes with Thermo Scientific Pierce Fast Blocking Buffer. The blots were probed with mixtures of mouse and rabbit primary antibodies, then with HRP-conjugated secondary antibodies. Wash steps were performed with the Fast Wash Buffer. Finally, blots were detected on film via Pierce ECL Western Blotting Substrate (Part No. 32106). HH3 = Histone H3; CycB = Cyclophilin B. |
