| Pulsar 650 (P-650) consists of a Ru(bpy)3+2 metal-ligand complex attached to a linker arm which is attached to a modified C nucleoside. The nucleoside is conjugated to CPG via a succinate linkage. Pulsar-650 CPG can be used for the synthesis of 3'-labeled oligonucleotides on any DNA synthesizer according to standard oligonucleotide synthesis procedures. Pulsar-650 has a very large Stokes’ shift with an absorption maximum at 450 nm and an emission maximum at 650 nm. Unlike most oligonucleotide dye labels, the Pulsar-650 label has a larger extinction coefficient at 260 nm than at the lambda max at 460 nm.
Pulsar 650 (P-650) is a fluorophore designed especially for use on the LightCycler® 1.2 and 2.0 real-time, qPCR instruments. In addition to its value in real-time PCR, Pulsar 650 is also useful for electrochemiluminescence, electrochemical detection, redox reactions, chemiluminescence and time-resolved luminescence.
LightCycler Users-
Users of the LightCycler 1.2 and 2.0 can now set up and run duplexed assays on your instrument by using two BHQ-quenched dual-labeled probes. Calibration is required for first time users. Additionally, LightCycler 2.0 users will need to spike FAM calibration dye into their Pulsar 650 capillaries. | - Synthesis conditions: Follow the instrument manufacturer's protocol when using this product.- Cleavage conditions:Use concentrated ammonia for 90 minutes at 25 °C when using standard amidites or 1:1 ammonia:methylamine (AMA) for 25 minutes at 25 °C when using fast deprotecting amidites.- Deprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF, G-PAC) use concentrated ammonia for 1 hour or AMA for 30 minutes at 60 °C. When using standard amidites (eg. C-Bz, G-iBu) use concentrated ammonia for 5 hours.- The mass this product adds after conjugation and work-up (the additional mass seen by mass spectrometry) is: 1116.13 |
