A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. It is reported that Siglec9 recognizes sialic acid in alpha2, 3-or alpha2, 6-glycosidic linkage to galactose. Using a specific antibody, Siglec9 was found to be expressed at high or intermediate levels by monocytes, neutrophils, and a minor population of CD16+, CD56- cells. Siglec9 also acts as an inhibitory receptor for MUC16 that attenuates T- andNK cell function.
