| Synthesis conditions:Use fast deprotecting amiditesCleavage conditions:Cleave in 2-Methoxyethylamine (Ald# 241067-50) and MeOH (1:3) for 3 hours at 25 °C or 1.5 hours at 60 °C.Deprotection conditions:Cleave and deprotect the CAL dye labeled oligo in 2-Methoxyethylamine (Ald# 241067-50) and MeOH (1:3) for 6 hours at 25 °C or 3 hours at 60 °C.
The use of strong bases, such as concentrated ammonia, causes degradation of dye and should be avoided.Due to the mild deprotection conditions we recommend using fast deprotecting amidites with this support. CAL Fluor dyes are light sensitive, prolonged exposure to light causes photobleaching.
