| Mixed base supports are used to create dNTP oligos used for mutation studies. The N Mix CPG (5'-DMT-[dA(Bz), dC(Ac), dG(DMF), T]-Gly-CPG) includes a mixture of bases A, C, G and T that have glycolate linkages to the CPG support. The glycolate linker allows for rapid cleavage of the oligonucleotides and is labile enough for base sensitive oligonucleotides. |

| Synthesis conditions:Follow the instrument manufacturer's protocol when using this product.Cleavage conditions:Use concentrated ammonia for 45 minutes at 25 °C or 1:1 ammonia:methylamine (AMA) for 15 minutes at 25 °C when using fast deprotecting amidites.Deprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF) use concentrated ammonia for 1 hour or AMA for 30 minutes at 60 °C.This support should be stored at -20°C. If the product is stored at higher temperatures for prolonged periods of time the glycolate linkage may cleave resulting in lower yields. The C- Acetyl group may also come off as a result of prolonged exposure to higher temperatures. This product is temperature sensitive. |
