产品详细信息Description: The MQ1-17H12 antibody reacts with human interleukin-2 (IL-2), a 17 kDa T cell growth factor and a major immunoregulatory cytokine. The MQ1-17H12 antibody is a non-neutralizing antibody.Applications Reported: This MQ1-17H12 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.Applications Tested: This MQ1-17H12 antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells. This can be used at 5 µL (0.5 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.eFluor® 450 is an alternative to Pacific Blue®. eFluor® 450 emits at 445 nm and is excited with the Violet laser (405 nm). Please make sure that your instrument is capable of detecting this fluorochome.Excitation: 405 nm; Emission: 445 nm; Laser: Violet Laser.Filtration: 0.2 µm post-manufacturing filtered.靶标信息Interleukin 2 (IL-2) is an immuno-modulatory cytokine that is important for the proliferation of activated T cells, differentiation of B cells, natural killer cells, monocytes and macrophages. IL-2 signals through the IL-2 receptor (IL-2R), a heterotrimeric protein complex whose gamma chain is also shared by interleukin 4 (IL-4) and interleukin 7 (IL-7). The expression of the IL-2 gene in mature thymocytes is monoallelic, which represents an unusual regulatory mode for controlling the precise expression of a single gene. The targeted disruption of a gene similar to IL-2 in mice leads to an ulcerative colitis-like disease that suggests an essential role of this gene in the immune response to antigenic stimuli.

| IL-2 Antibody (48-7029-42) in FlowNormal human peripheral blood cells were unstimulated (left) or stimulated for 5 hours with the Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975-03) (right) and then intracellularly stained with Anti-Human CD4 APC (Product # 17-0047-42) and Anti-Human IL-2 eFluor® 450, using the IC Fixation and Permeabilization Buffer Set (Product # 88-8824-00). Total viable cells, as determined by Fixable Viability Dye eFluor® 780 (Product # 65-0865-14), were used for analysis. |
