| Quasar® 705 is an indocarbocyanine which fluoresces in the red region of the visible spectrum and is a direct replacement for Cy™5.5 dye. Quasar 705 amidite is used for the 5' labeling of fluorogenic probes used in 5' nuclease assays, Molecular Beacons, Scorpion® primers, and other detection assays. This amidite does not contain a DMT protecting group and can only be added to the 5' terminus of the oligo. BHQ®-2 dye will quench the Quasar 705 moiety. |

| Synthesis conditions: Due to the mild cleavage and deprotection conditions, we recommend using fast deprotecting amidites. Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in sealed vial to ensure that all contents are dissolved. For optimal coupling efficiency, we recommend treating the amidite solution with 3-4 Å activated molecular sieves(10% v/v).Dilution: 100 µmol/1 mLDeprotection conditions: Oligonucleotides labeled with Quasar 705 may be deprotected using concentrated NH4OH for 8 hours at 25 °C or for 1 hour at 60 °C. Some degradation may occur when using NH4OH, the use of methylamine should be avoided.Quasar 705 is light sensitive material. Prolonged exposure to light may cause photo bleaching. This product is susceptible to oxidation when exposed to air or stored undessicated. Once product is dissolved in dry acetonitrile, use it immediately.
