TheMitoProbe™ DiIC1(5) Assay Kitprovides solutions of the membrane-potential-sensitive cyanine dye DiIC1(5), and a mitochondrial membrane-potential disrupter, CCCP, for the study of mitochondrial membrane potential. DiIC1(5) penetrates the cytosol of eukaryotic cells and accumulates primarily in mitochondria with active membrane potentials, producing bright, far-red fluorescence. DiIC1(5) staining intensity decreases with decreased mitochondrial membrane potential. |

图1.MitoProbe™ DiIC1(5) Assay Kit.MitoProbe™ DiIC1(5) Assay Kit Decrease in mitochondrial membrane potential as demonstrated with DiIC1(5) fluorescence due to the addition of carbonyl cyanide 3-chlorophenylhydrazone (CCCP). Jurkat cells (T-cell leukemia, human) were stained with 50 nM DiIC1(5) alone (blue line) or in the presence of 50 ?M CCCP (red line).图2.Multiparameter apoptosis assay.Control (top row) and 4-hour camptothecin-treated (bottom row) Jurkat cells were stained with 1 µM SYTOX® AADvanced™ Dead Cell Stain (S10349), 200 nM F2N12S (from A35137), and 50 nM MitoProbe™ DiIC1(5) (M34151). Dead cells were first excluded (histograms) by gating live cells (those that have lower SYTOX® AADvanced™ fluorescence, indicated by bars). Bivariant density plots show a two-parameter apoptosis assay for mitochondrial membrane potential loss (decreased DiIC1(5) fluorescence) and breakdown of membrane asymmetry (smaller F2N12S 585/530 nm fluorescence ratio). The A, L, and D labels on the graphs indicate apoptotic, live, and dead cells, respectively. |
