Secondary antibodies offer increased versatility enabling users to use many detection systems (e.g. HRP, AP, fluorescence). They can also provide greater sensitivity through signal amplification as multiple secondary antibodies can bind to a single primary antibody. Most commonly, secondary antibodies are generated by immunizing the host animal with a pooled population of immunoglobulins from the target species and can be further purified and modified (i.e. immunoaffinity chromatography, antibody fragmentation, label conjugation, etc.) to generate highly specific reagents.

Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody (R37121) in IF HeLa cells were transduced with CellLight® Nucleus-GFP (Product # C10602), grown over night. Following fixation, permeabilization and block using the Image-iT Fixation/Permeabilization Kit (Product # R37602), mMitochondria are labeled with mouse anti-ATP synthase subunit IF1 monoclonal antibody (Product # A-21355) and detected with ReadyProbes® Alexa Fluor® 594 Goat Anti-Mouse IgG Antibody (Product # R37121). Nuclei were stained with NucBlue® Fixed cell stain (Product # R37606). Images were acquired on the EVOS® FL Auto imaging system using a 20x objective (Product # AMEP4624). Mouse IgG (H+L) Cross-Adsorbed Secondary Antibody (R37121) in IFHeLa cells were transduced with CellLight® Tubulin-GFP (Product # C10508), grown over night, and fixed, permeabilized and blocked using the Image-iT Fixation/Permeabilization Kit (Product # R37602). Mitochondria were labeled with mouse anti-ATP synthase subunit IF1 monoclonal antibody (Product # A-21355) and detected with Alexa Fluor® 594 Goat Anti-Mouse IgG Antibody ReadyProbes® reagent (Product # R37121). Nuclei were stained with NucBlue Fixed (Product # R37606). Images were acquired on the EVOS® FL Auto imaging system using a 20x objective (Product # AMEP4624).
