DyLight 550 has an excitation/emission of 562/576 nm.Background/Target InformationThe 6x His tag is a synthetic oligo peptide consisting of 6 consecutive histidine residues (HHHHHH). The His tag is commonly expressed as a tag at either N-or C-terminal regions of recombinant proteins to allow isolation or purification by immobilized metal affinity chromatography. Epitope tagging is a technique in which a known epitope is fused to a recombinant protein using genetic engineering. By choosing a particular epitope and recombinant protein combination, epitope tagging makes it possible to detect proteins for which no antibody is available. His epitope tagged proteins that contain a stretch of histidine residues at the carboxyl or amino terminus are used for purification of recombinant proteins by means of metal chelate chromatography due to metal binding properties of histidine clusters on His tagged proteins. For example, the affinity of the histidine-tag motif to Ni2+ by chelation is strong and selective enough to enable purification of the protein to homogeneity by affinity chromatography on a Ni2+-NTA adsorbent. | 6x-His Tag Antibody (MA1-21315-D550) in IFImmunofluorescent analysis of HeLa cells transfected with a construct containing a 6x-His Epitope Tag. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA (Product # 37525) for 15 minutes at room temperature. Cells were probed with a DyLight 550-conjugated 6x-His Epitope Tag monoclonal antibody (Product # MA1-21315-D550) at a dilution of 1:25 for at least 1 hour at room temperature. Nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ArrayScan or ToxInsight Instrument at 20X magnification.6x-His Tag Antibody (MA1-21315-D550) in IFImmunofluorescence was performed on HeLa cells transfected with PTEN expressing pTT5 plasmid. 24 hours post transection, cells were fixed and permeabilized for detection of PTEN using Anti- 6x-His Epitope Tag, DyLight 550 Mouse Monoclonal Antibody (Product # MA1-21315D550, 5 µg/mL). Panel a) shows representative cells that were stained for detection and localization of PTEN protein (red), Panel b) is stained for nuclei (blue) using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Panel c) represents cytoskeletal F-actin staining using Alexa Fluor® 488 Phalloidin (Product # R415, 1:100). Panel d) is a composite image of Panels a, b and c clearly demonstrating cytoplasmic localization of PTEN. Panel e) shows no signal in untransfected cells.
