Product Specific Information33-9100 has been successfully used in ELISA, Immunofluorescence and Western Blot analysis of ZO-1.Target InformationZO-1 forms complexes with either ZO-2 or ZO-3. In addition, these proteins can also associate with claudin, occludin and F-actin, at tight junction stands, where they provide a linkage between the actin cytoskeleton and the tight junction. ZO-1 expression is significantly reduced in many breast cancer lines. ZO-2 and ZO-3 are ubiquitously expressed within epithelial tight junctions, and unlike ZO-1, which is also expressed at cell junctions of cardiac myocytes, ZO-2 is not expressed in nonepithelial tissue. ZO-1 is a protein located on a cytoplasmic membrane surface of intercellular tight junctions. ZO-1 may be involved in signal transduction at cell-cell junctions. Two transcript variants encoding distinct isoforms have been identified for the ZO-1 gene. ZO-1 Antibody (33-9100) in IFMDCK canine cells stained with Mouse anti-ZO-1 (Product # 33-9100) and Zymed's ZyMAX FITC-Goat Anti-Mouse IgG (H+L) (Product # 81-6511).ZO-1 Antibody (33-9100) in IFImmunofluorescent analysis of ZO-1 (green) in Caco-2 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes and blocked with 3% Blocker BSA (Product # 37525) in PBS for 15 minutes at room temperature. Cells were stained with or without ZO-1 mouse monoclonal antibody (Product # 33-9100), at a concentration of 5 µg/mL for 1 hour at room temperature, and then incubated with a Goat anti-Mouse IgG (H+L) Superclonal Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A28175) at a dilution of 1:1000 for 1 hour s at room temperature (both panels, green). Nuclei (both panels, blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ToxInsight at 20X magnification.ZO-1 Antibody (33-9100) in IFImmunofluorescent analysis of ZO-1 (red) and SOX2 (grey) in human iPSC-derived forebrain organoids derived at Day 40. The organoids were fixed with 4% PFA for 1 hour at room temperature, followed by incubation with 30% sucrose solution overnight at 4°C. The organoids were then embedded in OCT and cryosectioned at 5 µm, permeabilized with 0.2% Triton X-100 for 20 min, and blocked with 10% donkey serum in PBS for 30 min at room temperature. Organoid slices were stained with a Mouse ZO-1 monoclonal antibody (red; Product # 33-9100) at a dilution of 1:500, a Rabbit SOX2 polyclonal antibody (grey; Product # PA1-094X) at a dilution of 1:500, and a Chicken MAP2 polyclonal antibody (green) at a dilution of 1:1000 in blocking buffer overnight at 4°C, and then incubated with Donkey anti-Mouse Alexa Fluor 568 (Product # A10037), Donkey anti-Rabbit Alexa Fluor 647 (Product # A-31573), and Donkey Anti-Chicken Alexa Fluor 488 at a dilution of 1:1000 as well as DAPI (blue; 1:25000) in blocking solution at room temperature for 1 hour.
