ECL Western Blotting Substrate for detection of proteins on nitrocellulose or PVDFβ-actin and β-galactosidase protein in HeLa cell and E. coli lysates were detected by Western blotting. The membranes were blocked with 5% skim milk and probed with primary antibody at 1µg/mL. The membranes were washed and then incubated with 0.2µg/mL of HRP-conjugated goat anti-mouse IgG (Part No. 31430) and then washed again. Working solutions of the Pierce ECL Substrate and GE Healthcare ECL Reagent were prepared according to the manufacturers’ instructions and added to replicate membranes for 1 minute. The membranes were removed from the substrates and placed in plastic sheet protectors and exposed to Thermo Scientific CL-XPosure Film (Part No. 34090) and developed. |
