| CAL Fluor Red® 610 is an amidite that fluoresces in the orange-red region of the visible spectrum. CAL Fluor Red 610 amidite is used for the 5' labeling of fluorogenic probes used in 5' nuclease assays, Molecular Beacons, and other detection assays. This amidite does not contain a DMT protecting group and can only be added to the 5' terminus. BHQ®-2 dye will quench the CAL Fluor Red 610 moiety. The CAL Fluor Red 610 fluorophore is an alternative for Texas Red® dye. |

| Synthesis conditions: Due to the mild deprotection conditions we recommend using fast deprotecting amidites. Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in sealed vial to ensure that all contents are dissolved. We recommend using ethyl thiol tetrazole for the activator reagent with this product.Dilution: 100 µmol/mLDeprotection conditions:Cleave and deprotect the CAL dye labeled oligo, in the presence of the CPG support, in 2-Methoxyethylamine(Ald# 241067-50) and MeOH (1:3) 3 hours at 60 °C. Filter sample and evaporate supernatant. Re-dissolve the CAL dye labeled oligo in water.The use of strong bases, such as concentrated ammonia, causes degradation of the dye and should be avoided.CAL Fluor Red 610 is light sensitive. Prolonged exposure to light may cause photo bleaching. This product is susceptible to oxidation when left out in the open or undessicated. Unused amidite solution can be stored under argon and its functionality maintained for up to 24 hours.
