| Pulsar® 650 (P-650) is a new fluorophore designed especially for use on the LightCycler® 1.2 and 2.0 real-time, qPCR instruments. In addition to its value in real-time PCR, Pulsar 650 is also useful for electrochemiluminescence, electrochemical detection, redox reactions, chemiluminescence and time-resolved luminescence. CPG-derivatized with P-650 is used for the synthesis of 3'-labeled oligonucleotides on any DNA synthesizer according to standard oligonucleotide synthesis procedures.
LightCycler Users-
Users of the LightCycler 1.2 and 2.0 can now set up and run duplexed assays on your instrument by using two BHQ®-quenched dual-labeled probes. Calibration is required for first time users. Additionally, LightCycler 2.0 users will need to spike FAM™ calibration dye into their Pulsar 650 capillaries. | Synthesis conditions:Follow the instrument manufacturer's protocol when using this product.Cleavage conditions:Use concentrated ammonia for 90 minutes at 25 °C when using standard amidites or 1:1 ammonia:methylamine (AMA) for 25 minutes at 25 °C when using fast deprotecting amidites.Deprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF, G-PAC) use concentrated ammonia for 1 hour or AMA for 30 minutes at 60 °C. When using standard amidites (eg. C-Bz, G-iBu) use concentrated ammonia for 5 hours.Fluorescent lifetime: ca.1 microsecond Users of the LightCycler 1.2 and 2.0 can now set up and run duplexed assays on your instrument by using two BHQ-quenched dual-labeled probes as follows:
Probe 1: 5' FAM/3' BHQ-1 for detection in the 530 nm (F1) channel
Probe 2: 5' BHQ-2/3' Pulsar-650 for detection in the 705 nm (F3) channel
LightCycler 1.2 Users-to successfully duplex P-650 and FAM probes, a universal Color Compensation file (CC file) must be downloaded and installed onto your hard drive. This file instructs the LightCycler 1.2 to subtract any crosstalk from these reporters outside their designated channels. Instructions for calibration and a CC File, created by our partner Fluoresentric (www.fluoresentric.com), are available for download at no charge from the Biosearch website at www.btidna.com/download/calibration.asp
LightCycler 2.0 Users- Because the LightCycler 2.0 seeks FAM fluorescence from each capillary during the optics optimization, FAM calibration dye needs to be spiked into any capillary that is lacking a FAM probe, including Pulsar 650 singleplex reactions. We recommend spiking the calibration dye to a final concentration of 10 nM. Please inquire about the availability of this calibration dye. To successfully duplex P-650 and FAM probes an experimental file (.ixo) needs to be imported into your LightCycler software (version 4.0). This file instructs the LightCycler to subtract any crosstalk from these reporters outside their designated channels, and is available for download at no charge from the Biosearch website at www.btidna.com/download/calibration.asp
As alternatives to using the provided files, CC objects can also be generated using TaqMan® probes themselves or T10 calibration dyes (inquire about availability). Detailed instructions for all three of these methods are available at the above web address.
