靶标信息Cytokeratins (CK) are intermediate filaments of epithelial cells, both in keratinizing tissue (ie. skin) and non-keratinizing cells (ie. mesothelial cells). At least 20 different CKs can be identified. Biochemically, most members of the CK family fall into one of two classes, type I (acidic polypeptides) and type II (basic polypeptides). Belonging to the type I subfamily of low molecular weight keratins and existing in combination with keratin 5, keratin 14 distinguishes stratified epithelial cells from simple epithelial cells and is useful in identification of squamous cell carcinomas. Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C) incubated with KRT14 polyclonal antibody (Product # PA5-99310) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 at a dilution of 1:600. KRT14 Antibody (PA5-99310) in IHC (P)Immunohistochemistry analysis of paraffin-embedded KRT14 in human colon cancer tissues sections. Antigen retrieval was performed using citrate buffer. Samples were blocked with blocking buffer (1.5 hr, 22°C), incubated with KRT14 polyclonal antibody (Product # PA5-99310) using a dilution of 1:100 (1.5 hr, 22°C), followed by HRP conjugated goat anti-rabbit.
