H3 along with four core histone proteins binds to DNA forming the structure of the nucleosome. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. Post translationally, histones are modified in a variety of ways to either directly change the chromatin structure or allow for the binding of specific transcription factors. The N-terminal tail ofhistone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

H3K9me2 Antibody (710815) in IFFor immunofluorescence analysis HeLa cells were fixed and permeabilized for detection of H3K9me2 using Histone H3K9me2 Recombinant Rabbit Polyclonal Antibody (Product # 710815, 2 µg/mL) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 1:2000). Panel a) shows representative cells that were stained for detection and localization of H3K9me2 protein (green), Panel b) is stained for nuclei (blue) using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). Panel c) represents cytoskeletal F-actin staining using Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of H3K9me2. Panel e) shows loss of signal by competition with the H3K9me2 peptide, demonstrating antibody specificity, and panel f) demonstrates no competition with the non-methylated peptide. H3K9me2 Antibody (710815) in ELISACross-reactivity ELISA for Anti-Histone H3K9me2 Recombinant Rabbit Polyclonal Antibody (Product # 710815) was performed against H3K9me2, H3K9 Nme, H3K9me3 and H3K9me1 by coating respective antigens at 1 µg/mL. Antibodies at 10, 2.5, 0.625 and 0.156 µg/mL were added and detected using Goat anti-Rabbit IgG (H+L) Secondary Antibody, HRP conjugate (Product # G-21234, 1:5000 dilution). The plate was developed using Stabilized Chromogen, TMB (Product # SB02).
