靶标信息The transcription factor ATF-2 (also called CRE-BP1) binds to both AP-1 and CRE DNA response elements and is a member of the ATF/CREB family of leucine zipper proteins. ATF-2 interacts with a variety of viral oncoproteins and cellular tumor suppressors and is a target of the SAPK/JNK and p38 MAP kinase signaling pathways. Various forms of cellular stress, including genotoxic agents, inflammatory cytokines and UV irradiation, stimulate the transcriptional activity of ATF-2. Cellular stress activates ATF-2 by phosphorylation of Thr69 and Thr71. Both SAPK and p38 MAPK have been shown to phosphorylate ATF-2 at these sites in vitro and in cells transfected with ATF-2.

Samples were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, blocked with 10% serum (45 min at 25°C), incubated with mouse anti-beta tubulin and Phospho-ATF2 (Thr69, Thr51) polyclonal antibody (Product # PA5-104804) using a dilution of 1:200 (1 hr, 37°C), and followed by goat anti-rabbit IgG Alexa Fluor 594 (red) and goat anti-mouse IgG Alexa Fluor 488 (green). Phospho-ATF2 (Thr69, Thr51) Antibody (PA5-104804) in WB Western blot analysis of Phospho-ATF2 (Thr69, Thr51) in various samples (Lane 1: Heat-shock treated MCF7 cells, blocked with antigen-specific peptides, Lane 2: Heat-shock treated MCF7 cells, Lane 3: Heat-shock treated HeLa cells).
