产品详细信息It is not recommended to aliquot this antibody.This antibody is not cross-reactive with the related GCN5L2 protein. 靶标信息PCAF (histone acetyltransferase KAT2B) functions as a histone acetyltransferase (HAT) to promote transcriptional activation. It has significant histone acetyltransferase activity with core histones (H3 and H4) and nucleosome core particles. PCAF also acetylates non-histone proteins such as ACLY, PLK4, and TBX5. It inhibits cell-cycle progression and counteracts the mitogenic activity of the adenoviral oncoprotein E1A. PCAF also acts as a circadian transcriptional coactivator which enhances the activity of the circadian transcriptional activators NPAS2-ARNTL/BMAL1 and CLOCK-ARNTL/BMAL1 heterodimers. It is involved in heart and limb development by mediating acetylation of TBX5 and acetylation regulating nucleocytoplasmic shutting of TBX5.

PCAF Antibody (MA5-11186) in WB Western blot was performed using PCAF Monoclonal Antibody (E.107.0) (Product # MA5-11186) and a 93 kDa band corresponding to PCAF was observed in all the cell lines tested. Whole cell extracts (30 µg lysate) of HeLa (Lane 1), PC-3 (Lane 2), C2C12 (Lane 3), HEK-293 (Lane 4) and (45 µg lysate) of LNCAP (Lane 5) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001).The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). PCAF Antibody (MA5-11186) in WB Knockdown of PCAF was performed by transfecting LNCaP cells with PCAF specific siRNAs (Silencer® select Product # s16894). Western blot analysis (Fig. a) was performed using whole cell extracts from the PCAF knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with PCAF Monoclonal Antibody (E.107.0) (Product # MA5-11186, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to PCAF. An uncharacterized band (*) was observed at ~42 kDa.
