This protein methylates 'Lys-9' of histone H3 in vitro. H3 'Lys-9' methylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 proteins to methylated histones. EHMT1 is Probably targeted to histone H3 by different DNA-binding proteins like E2F6, MGA, MAX and/or DP1. During G0 phase, it probably contributes to silencing of MYC- and E2F-responsive genes, suggesting a role in the G0/G1 transition of the cell cycle.

EHMT1 Antibody (A301-642A) in IHCDetection of human EHMT1 by immunohistochemistry. Sample: FFPE section of human breast carcinoma. Antibody: Affinity purified rabbit anti-EHMT1 (Cat. No. A301-642A) used at a dilution of 1:200 (1µg/ml). Detection: DAB. EHMT1 Antibody (A301-642A) in WBDetection of human EHMT1 by western blot and immunoprecipitation. Samples: Whole cell lysate (5, 15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-EHMT1 antibody A301-642A used for WB at 0.04 µg/ml (A) and 0.4 µg/ml (B) and used for IP at 3 µg/mg lysate. EHMT1 was also immunoprecipitated by rabbit anti-EHMT1 antibody A301-643A, which recognizes a downstream epitope. Detection: Chemiluminescence with exposure times of 10 seconds (A) and 30 seconds (B).
