Target InformationSSEA-4 (Stage-specific embryonic antigen 4) is a cell surface antigen, expressed along with SSEA-3, TRA-1-60 and TRA-1-81 in embryonic stem cells, embryonal carcinoma cells and induced pluripotent stem cells (iPS). In human cells, these surface markers are down-regulated during the differentiation process, while SSEA-1 is absent in undifferentiated human stem cells and upregulated on the cell surface after retinoic acid mediated differentiation. Conversely, in mouse stem cells, SSEA-1 expression decreases with differentiation while SSEA-3 and SSEA-4 expression increase.For Research Use Only. Not for use in diagnostic procedures. SSEA4 Antibody (MA1-021) in IFImmunofluorescent analysis of SSEA4 using anti-SSEA4 monoclonal antibody (Product # MA1-021) shows expression in human teratocarcinoma NCCIT cells (shown in green) but not in negative control HeLa cells. Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature. Cells were blocked with 1% Blocker BSA (Product # 37525) for 15 minutes at room temperature. Cells were probed with a mouse monoclonal antibody recognizing SSEA4 (Product # MA1-021), at a dilution of 1:50 for at least 1 hour at room temperature. Cells were washed with PBS and incubated with DyLight 488 goat-anti-mouse secondary antibody (Product # 35503) at a dilution of 1:400 for 30 minutes at room temperature. Nuclei (blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken on a Thermo Scientific ArrayScan at 20X magnification. SSEA4 Antibody (MA1-021) in FlowFlow cytometry analysis of SSEA4 on human Gibco® Episomal iPSC line (Product # A18945) grown on vitronectin in Essential 8™ medium (Product # A1517001). Cells were fixed, permeabilized, and then stained with a 1:50 dilution of SSEA4 mouse monoclonal antibody (Product # MA1-021x) or 10 µg/mL of CD44 mouse monoclonal antibody (negative control, Product # MA5-13890) in 5% FBS/PBS. After incubation of the primary antibody for 1 hour on ice, the cells were stained with a DyLight 488-conjugated goat anti-mouse IgG secondary antibody (Product # 35502) at a dilution of 1:500 for 1 hour on ice. A representative 10,000 cells were acquired for each sample. SSEA4 Antibody (MA1-021) in IFImmunofluorescent analysis of SSEA-4 using anti-SSEA-4 monoclonal antibody (Product # MA1-021) shows staining on the cell surface of human HEL 11.4 iPS cells, indicating pluripotency. SSEA-4 staining (green) and an overlay image of SSEA-4 with DAPI (blue) is shown. HEL 11.4 iPS cells were grown on matrigel coated chamber slides and fixed with formaldehyde prior to staining. Cells were probed with a Monoclonal antibody recognizing SSEA-4 (Product # MA1-021) at a dilution of 1:100 overnight at 4 °C, washed with PBS and incubated with a fluorescein conjugated secondary antibody at a dilution of 1:100 for 60 minutes at room temperature. Images were taken at 20X magnification.
