The exact population can be produced in every lot, circumventing the biological variability typically associated with polyclonal antibody production.Target InformationRex1 (zinc finger protein 42 homolog (ZFP42)) is a member of a class of DNA binding proteins which act as transcriptional regulators of other genes. Specifically, Rex1 is involved in the reprogramming of X-chromosome inactivation during the acquisition of pluripotency. It is required for efficient elongation of TSIX – a non-coding RNA antisense to XIST. Rex1 binds the DXPas34 enhancer within the TSIX promoter. It is also involved in ES cell self-renewal. Rex1 is expressed in healthy kidney, epidermal keratinocytes, prostate epithelial cells, bronchial/small airway lung epithelial cells, and embryonic stem cells. Rex1 Antibody (710190) in WBWestern blot analysis of Rex1 was performed by loading 20 µg of HeLa, 3T3, NRK, MDCK, and Cos7 whole cell lysates and 5uL of PageRuler Plus Prestained Protein Ladder (Product # 26619) per well onto a 4-20% Tris-Glycine polyacrylamide gel. Proteins were transferred to a nitrocellulose membrane using the G2 Blotter (Product # 62288), and blocked with 5% milk in TBST for 1 hour at room temperature. Rex1 was detected at ~35 kD using a Rex1 recombinant rabbit oligoclonal antibody (Product # 710190) at a concentration of 2 µg/mL in 5% milk in TBST overnight at 4C on a rocking platform, followed by a Goat anti-Rabbit IgG-HRP Superclonal™ secondary antibody (Product # A27036) at a concentration of 1 µg/mL for at least 30 minutes at room temperature. Chemiluminescent detection was performed using SuperSignal West Pico substrate (Product # 34080).

Rex1 Antibody (710190) in IFImmunofluorescent analysis of Rex1 in HeLa cells using a Rex1 recombinant rabbit oligoclonal antibody (Product # 710190) followed by detection using an Alexa Fluor 488-conjugated goat anti-rabbit secondary antibody (green) (Image A). Nuclei were stained using DAPI (Image B) and actin stained with Alexa Fluor 594 phalloidin (red) (image C). Image D is a composite image showing nuclear localization of Rex1.

Rex1 Antibody (710190) in ChIPChromatin immunoprecipitation analysis of Rex1 performed using cross-linked chromatin from 1 x 10^6 HCT116 human colon carcinoma cells treated with serum for 0, 15, and 60 minutes. Immunoprecipitation was performed using a multiplex microplate Matrix ChIP assay (see reference for Matrix ChIP protocol: http://www.ncbi.nlm.nih.gov/pubmed/22098709) with 1.0 µL/100 µL well volume of a Rex1 rabbit oligoclonal antibody (Product # 710190). Chromatin aliquots from ~1 x 10^5 cells were used per ChIP pull-down. Quantitative PCR data were done in quadruplicate using 1 µL of eluted DNA in 2 µL SYBR real-time PCR reactions containing primers to amplify -15kb upstream of the human Egr-1 locus, or exon-1 or exon-2 of Egr-1. PCR calibration curves were generated for each primer pair from a dilution series of sheared total genomic DNA. Quantitation of immunoprecipitated chromatin is presented as signal relative to the total amount of input chromatin. Results represent the mean +/- SEM for three experiments.
