大鼠IF中的IgG（H + L）交叉吸附二抗（A-21210）兔抗大鼠IgG（H + L）二抗Alexa Fluor 488偶联物的免疫荧光分析是使用用α微管蛋白（YL1 / 2）大鼠单克隆抗体（产品编号MA1-80017）染色的A549细胞进行的。细胞用4％多聚甲醛固定10分钟，用0.1％Triton™X-100透化10分钟，用1％BSA封闭1小时，并在室温下用2 µg / mL大鼠一抗标记3小时。在室温下，将兔抗大鼠IgG（H + L）二抗Alexa Fluor 488偶联物（产品编号A-21210）以0.5μg/ mL的浓度在含0.2％BSA的磷酸盐缓冲液中使用45分钟，检测细胞质中的α微管蛋白（面板a：绿色）。在SlowFade®Gold Antifade Mountant（产品编号S36938）中，用DAPI将细胞核（面板b：蓝色）染色。 F-肌动蛋白用若丹明鬼笔环肽（产品编号R415，1：300）染色（图c：红色）。面板d代表合成图像。单独的二抗（图f）或同种型对照（图e）未观察到非特异性染色。图像以60倍放大率拍摄。
