A target-specific antibody is coated to the bottom of the wells of a microplate, which is an overnight process. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. A sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. IL-23 interacts with IL-12Rbeta1 and a beta2-like receptor subunit with STAT4 binding domain, IL-23R. IL-23 is secreted by activated mouse and human dendritic cells and is involved in the inflammation process and associated with autoimmune diseases. Mouse IL-23 was found not to induce significant amounts of IFN-gamma, induces strong proliferation of memory T cells (but not naive T cells), and activate mouse memory T cells to produce the proinflammatory cytokine IL-17. On the other hand, human IL-23 induces proliferation of memory T cells, induces moderate levels of IFN-gamma, and is produced by naive and memory T cells. IL-23 plays an important role in stimulating memory T-cells, and is required for the induction, expansion, maintenance, and downstream effector functions of Th17 cells, which play a vital role in upregulating neutrophil chemokines and various pro-inflammatory cytokines. IL-23 interacts with the receptors IL-12R beta1 and IL-23R to activate the Jak-Stat signaling cascade.
