| 3'-DMT-dC(Ac) Super Column; 1000 Å is packed with 3'-DMT-dC(Ac)-Suc-CPG, an inverse linkage CPG used to create a 3'-3' linkage on the end of an oligonucleotide. An inverted 3'-3' linkage is desirable when needing to block polymerase extension through the 3'-terminus of a PCR probe, additionally oligonucleotides with inverted 3'-linkages are used for some sequencing techniques. The 1000 Å pore size is best suited for the synthesis of DNA sequences up to 100-mers in length. |

| Protecting group deblock conditions:3'DMT groups detritylate slowly. Increase detritylation time by 50% for 1st tritylCleavage conditions:Use concentrated ammonia for 90 minutes at 25 °C when using standard amidites or 1:1 ammonia:methylamine (AMA) for 25 minutes at 25 °C when using fast deprotecting amidites.Deprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF) use concentrated ammonia for 1 hour or AMA for 30 minutes at 60 °C. When using standard amidites (eg. C-Bz, G-iBu) use concentrated ammonia for 5 hours at 60 °C.
