By reducing the distance over which diffusion needs to occur, the time required for sample molecules to interact with interior binding sites is also reduced. Diffusion is no longer limiting and flow rates can be dramatically increased—without compromising resolution or capacity. Dynamic binding capacity over a range of operating flow ratesPOROS 50 HQ and XQ provide high dynamic binding capacities and excellent capture efficiencies at higher linear velocities. Dynamic binding capacity was determined by loading a 10 mg/mL BSA solution onto a 3.3 mL until a 5% breakthrough was reached. The experiment was then repeated using different flow rates. Experimental conditions: Column: 0.46 cmD x 20 cmL; Load condition: 10 mg/mL BSA in 20 mM Tris, pH 8.0 Resolution over a range of operating flow ratesPOROS 50 HQ is capable of achieving excellent seperation using a very steep gradient and maintaining resolution at multiple flow rates. A protein mixure of holo-transferrin, ovalbumin, and soybean trypsin inhibitor was loaded onto a 3.3 mL column and then eluted using a 0-50% B gradient over 10 column volumns. The experiment was then repeated using different flow rates. Experimental conditions: Column: 0.46 cmD x 20 cmL, Buffer A: 20 mM Tris, pH 8.0, Buffer B: 20 mM Tris, 1.0 M NaCl, pH 8.0. |
