They undergo various post-translational modifications including methylation, acetylation and phosphorylation to facilitate chromatin regulation. Modified histones display varied distribution patterns at the heterochromatin and the euchromatin, hence defining actively transcribed or repressed genes in the eurochromatin and the 'silenced' loci in the heterochromatin. These modifications in turn serve as epigenetic markers for transcriptional status of a gene and landing sites for transcriptional complexes, fundamental to understanding several biological processes.

Histone H3 Antibody (702023) in IFImmunofluorescence was performed on fixed and permeabilized MCF7 cells for detection of Histone H3 using ABfinity™ Anti-Histone H3 (N-term) Recombinant Rabbit Monoclonal Antibody (Product # 702023, 1 µg/mL) and labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034, 0.4 µg/mL, 1:2500). Panel a) shows representative cells that were stained for detection and localization of Histone H3 protein (green), Panel b) is stained for nuclei (blue) using SlowFade® Gold Antifade Mountant with DAPI (Product # S36938, 1:50). Panel c) represents cytoskeletal F-actin staining using Alexa Fluor® 594 Phalloidin (Product # A12381, 1:200). Panel d) is a composite image of Panels a, b and c clearly demonstrating nuclear localization of Histone H3. Panel e) represents control cells with no primary antibody to assess background. Histone H3 Antibody (702023) in WBWestern blot analysis was performed on acid extracts (20µg lysate) of HEK-293 (Lane 1), HEL 92.1.7 (Lane 2), K-562 (Lane 3), PC-12 (Lane 4) and THP-1 (Lane 4). The blot was probed with ABfinity™ Anti-Histone H3 (N-term) Recombinant Rabbit Monoclonal Antibody (Product # 702023, 2µg/mL) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25µg/mL, 1:4000 dilution). A 17 kDa band corresponding to Histone H3 was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE®12 % Bis-Tris gel (Product # NP0342BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).
