OCT4 Antibody (MA1-104-D488) in IF

Immunofluorescent analysis of Oct4/Pou5F1 (green) shows nuclear staining in human HEL 11.4 iPS cells, indicating pluripotency. Oct4/Pou5F1 staining (left panel), DAPI staining (middle panel) and an overlay image of Oct4/Pou5F1 with DAPI (right panel) are shown. HEL 11.4 iPS cells were grown on matrigel-coated chamber slides and fixed with 4% formaldehyde for 15 minutes prior to staining. Cells were probed with a DyLight 488-conjugated Oct4/Pou5F1 monoclonal antibody (Product # MA1-104-D488) in 0.1% Tween-PBS at a dilution of 1:20, and incubated overnight at 4C. Images were taken at 20X magnification.

Immunofluorescence analysis of OCT-4 was performed using 70% confluent log phase NTERA-2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with OCT-4 Mouse Monoclonal Antibody (Product # MA1-104-D488) at 5µg/ml in 0.1% BSA, incubated at 4 degree celsius overnight (Panel a: green). Nuclei (Panel b: blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). Panel d represents the merged image showing nuclear localization. Panel e shows OCT-4 negative cell line HeLa with no signal. Panel f represents control cells with Isotype control to assess background. The images were captured at 60X magnification.

Antibody specificity was demonstrated by detection of differential basal expression of the target across cell models owing to their inherent genetic constitution. Immunofluorescence analysis using OCT-4 Monoclonal Antibody (Product # MA1-104-D488) showed expression of OCT-4 in NTERA-2 and not in HeLa which is reported to be negative model for OCT-4 expression.
