The product of the SOX2 gene is required for stem-cell maintenance in the central nervous system, and also regulates gene expression in the stomach. The SOX2 gene lies within an intron of another gene called SOX2 overlapping transcript (SOX2OT). Further, SOX2 protein may act as a transcriptional activator after forming a protein complex with other proteins. Mutations in the SOX2 gene have been associated with bilateral anophthalmia, a severe form of structural eye malformation, optic nerve hypoplasia and syndromic microphthalmia.For Research Use Only. SOX2 Antibody (MA1-014-D488) in IFImunofluorescent analysis of SOX2 (green) in Human episomal iPSC cells. The cells were fixed with formalin for 15 minutes, permeabilized with 0.1% Triton X-100 in TBS for 10 minutes, and blocked with 3% Blocker BSA (Product # 37525) for 30 minutes at room temperature. Cells were stained with Dylight 488-conjugated SOX2 monoclonal antibody (right, Product # MA1-014-D488) or Dylight 488-conjugated Mouse IgG1 isotype control antibody (left, Product # MA1-191-D488), at a dilution of 1:50 and incubated for 1 hour at room temperature. Nuclei (both panels, blue) were stained with Hoechst 33342 dye (Product # 62249). Images were taken at 20X magnification.SOX2 Antibody (MA1-014-D488) in IFImmunofluorescent analysis of SOX2 (green) shows nuclear staining in human HEL 11.4 iPS cells, indicating pluripotency. SOX2 staining (left panel), DAPI staining (middle panel) and an overlay image of SOX2 with DAPI (right panel) are shown. HEL 11.4 iPS cells were grown on matrigel-coated chamber slides and fixed with 4% formaldehyde for 15 minutes prior to staining. Cells were probed with a DyLight 488-conjugated SOX2 monoclonal antibody (Product # MA1-014-D488) in 0.1% Tween-PBS at a dilution of 1:20, and incubated overnight at 4C. Images were taken at 20X magnification.SOX2 Antibody (MA1-014-D488) in IFImmunofluorescent analysis of SOX2 (green) showing nuclear staining of NCCIT cells (right panel) compared to negative HeLa cell control (left panel). The cells were fixed with formalin for 15 minutes, permeabilized with 0.1% Triton X-100 in TBS, washed, and then blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a DyLight 488-conjugated SOX2 monoclonal antibody (Product # MA1-014-D488) in 3% BSA-PBS at a dilution of 1:100 and incubated for 1 hour at 37C in the dark. F-Actin (left panel, red) was stained with DyLight 554 Phalloidin (Product # 21834) and nuclei (left panel, blue) were stained with DAPI. Images were taken at 60X magnification.
