Secondary antibodies offer increased versatility enabling users to use many detection systems (e.g. HRP, AP, fluorescence). They can also provide greater sensitivity through signal amplification as multiple secondary antibodies can bind to a single primary antibody. Most commonly, secondaryantibodies are generated by immunizing the host animal with a pooled population of immunoglobulins from the target species and can be further purified and modified (i.e. immunoaffinity chromatography, antibody fragmentation, label conjugation, etc.) to generate highly specific reagents.

Mouse IgG (H+L) Secondary Antibody (A28183) in WBEndogenous alpha-tubulin (a), beta-actin (b) and GAPDH (c) was detected at 52 KDa, 46 KDa and 37 KDa respectively using mouse anti-alpha-tubulin (Product # 32-2500), beta-actin (Product # AM4302) and mouse anti-GAPDH (Product # 39-8600) primary antibody and and Goat anti-mouse IgG (H+L) Superclonal Secondary Antibody, Alexa Fluor 680 Conjugate (Product # A28183) (1 µg/mL 1:1000 dilution). Western blot analysis was performed on whole cell extracts (20 µg lysate) of HeLa human cervical carcinoma cells (lane 1-5) using the Xcell Surelock Electrophoresis system and iBlot® Dry Blotting System.
