Click-iT HPG kits are used to detect newly synthesized proteins.Inhibition of the protein decreases the Click-iT HPG signal in U2- OS cells. The graph above quantifies minimum and maximum for both color variants (the Alexa Fluor® 488 and Alexa Fluor 594kits); also shown are the images for vehicle and compound. Detection of protein synthesis using the Click-iT HPG Alexa Fluor 594 Protein Synthesis Kit.U2-OS cells were cultured in the presence of 50uM HPG for one hour followed by 50um ManNaz for 24 hours. Cells were then fixed and permeabilized. HPG was detected using the Click-iT HPG Alexa Fluor 594 Kit. ManNaz was detected using an Alkyne modified Alexa Fluor 488 dye. |
