| The Image-iT® LIVE Mitochondrial Transition Pore Assay Kit provides a proven method[REF PN36389 ,PN32679] of measuring mitochondrial permeability transition pore opening. The kit includes a dye and a quencher that are easily loaded into cells. In healthy cells, the dye, but not the quencher, enters mitochondria. The mitochondria remain brightly fluorescent until mitochondrial pore activation permits quenching of the fluorescence. |

Cells are counterstained with MitoTracker® Red CMXRos (Cat. no. M7512) to show mitochondria and with Hoechst 33342 (Cat. nos. H1399, H3570, H21492) to show nuclei. Panel A shows the uniform cellular fluorescence from unquenched calcein (Cat. nos. C1430, C3100MP). Panel B shows the mitochondrial pattern after adding cobalt. Panel C shows the loss of calcein fluorescence with the addition of ionomycin (mitochondria still visible from MitoTracker® red). Cyclophilin D activity is necessary for MPTP formation, and is inhibited by cyclosporin A. Consequently, inhibition of pore formation by cyclosporin A has been used as an argument that a function is MPTP-specific.1,2 Panel D shows that the calcein pattern is retained when cyclosporin A is added before to ionomycin, indicating that the ionomycin-triggered change observed here is an MPTP-mediated event. |
